در پژوهش حاضر، سینتیک و مکانیسم واکنش اسید گالیک و متیل گالات در مهار رادیکال DPPH● در طیفی از حلال های شیمیایی مورد مطالعه قرار گرفت. برای این منظور، پارامترهای سینتیکی از جمله ثابت سرعت ظاهری، ثابت سرعت مرتبه دوم و توانایی دادن هیدروژن آنتی اکسیدان و سایر پارامترهای مربوط به آزمون DPPH● مانند دسترسی فضایی به جایگاه واکنش رادیکال، غلظت موثره (IC50) و استوکیومتری واکنش محاسبه شدند. کاهش میزان جذب تابشی رادیکال DPPH● در محدوده 517 تا 520 نانومتر در غلظت ثابت DPPH● (حدود 60 میکرومولار) و غلظت متغیر آنتی اکسیدان پایش شد. تفاوت معنی دار مقادیر ثابت سرعت مرتبه دوم در حلال های الکلی نسبت به سایر حلال ها نشان داد حلال قادر است مکانیسم واکنش را از مسیر اصلی انتقال اتم هیدروژن (HAT) به مکانیسم انتقال الکترون (SPLET) تغییر دهد. این حاکی از پشتیبانی حلال از یونیزاسیون آنتی اکسیدان و یا حمله نوکلئوفیلی و احیاء مجدد ساختمان شیمیایی آنتی اکسیدان است. به کمک بررسی سرعت اولیه واکنش و نسبت اختلاف غلظت DPPHf/ΔDPPHiΔ در خصوص هر آنتی اکسیدان، واکنش اسید گالیک و متیل گالات با DPPH● به سه گروه سریع، متوسط و آهسته تقسیم بندی شد. همچنین نشان داده شد میزان دسترسی فضایی و اثر ساختمان شیمیایی آنتی اکسیدان و حلال بر نزدیک شدن آنتی اکسیدان به محل جایگاه واکنش رادیکال، سینتیک واکنش را به طرز مشهودی تحت تاثیر قرار می دهند. سرانجام، مشخص گردید متیل گالات نسبت به اسید گالیک حائز نتایج ضعیف تری از نظر غلظت موثره و استوکیومتری واکنش است که این ممکن است به آزادی فضایی و دسترسی بهتر اسید گالیک به جایگاه واکنش رادیکال DPPH● مربوط بوده باشد.

In this research, the kinetics and mechanism of the reaction between the DPPH● radical and two antioxidant compounds (gallic acid and methyl gallate) were investigated in different solvents. For this purpose, kinetic parameters such as pseudo-first-order rate constant, second-order rate constant, hydrogen bond donor ability of antioxidant, and some other parameters related to DPPH● test such as steric accessibility to the DPPH● radical site, IC50, the stoichiometry of the reaction were calculated. Studies were performed by monitoring the decrease of DPPH● absorbance in the range of 517-520 nm, at constant DPPH● concentration (~60 μM) and variable concentrations of antioxidants. The values showed a significantly higher second-order rate constant in alcoholic solvents, indicating that the kinetic solvent effects may lead to changes in the reaction mechanism from the main hydrogen atom transfer (HAT) to electron transfer (SPLET), which may be related to the solvent properties such as supporting the antioxidant ionization or regeneration of antioxidant structure by a nucleophilic attack. Exploiting the initial reaction rate and the ΔDPPHf/ΔDPPHi ratio in each antioxidant, gallic acid and methyl gallate reactions with DPPH● could be divided into three groups: rapid, medium, and slow. The results also showed that the steric accessibility as well as the effect of antioxidant structure and solvent type on the antioxidant closeness to the radical reaction site affect the reaction kinetics dramatically. Finally, methyl gallate showed weaker results than gallic acid regarding the IC50 and reaction stoichiometry, which this can be related to the steric freedom and better access of gallic acid to the radical reaction site.
